The overcoming of tumor antigen heterogeneity in diffuse midline glioma (DMG) with help from next generation CAR T-cell therapy<\/h2><\/div>

In this project we want to develop a new type of CAR T-cell therapy, which delivers a number of bi specific antibodies aimed at tumor invasive macrophage\/microglia cells in the tumor environment. In doing this we can aim the attack on the tumor cells with several antigens at once and potentially overcome problems with tumor antigen heterogeneity.<\/h4><\/div>

The evaluation of the response from different cellular populations targeted drug screening for brain tumors in children to identify new drug combinations<\/h2><\/div>

The target with this study is to identify markers for several tumor cell populations primarily for DMG and medulloblastoma, which can be used with flow cytometry to evaluate how both standard treatment and the targeted treatment affect the different tumor cell populations. By discovering drugs that target single cellular populations, do we want to examine combinations of drugs that target several cell populations at the same time. Our hypothesis is that by targeting several tumor cell populations, we can develop more effective treatments, which prevents tumor cells from growing back.<\/h4><\/div>

Protein, sugar, and fat burning in children and adolescents with CNS-tumors before and after treatment<\/h2><\/div>

The project aims to describe the changes that happens in the metabolism more specific the metabolic conversion of protein, sugar, and fat in the approx. two-year long treatment period, that the children go through. This will contribute to a better understanding of the illness and promote the development of new treatments, that prevent malnutrition, loss of muscle mass and lifestyle diseases. The project includes children and adolescents within the age range of 2-21 years who is either newly diagnosed with a brain tumor or done with their treatment for a brain tumor within the last month.<\/h4><\/div>

Trombocytter akkumulerer RNA fra tumorer. Det er vist hos voksne, at man ved kortl\u00e6gning af mRNA indhold i trombocytter kan kortl\u00e6gge specifikke genekspressionsprofiler og ogs\u00e5 identificerer visse af tumorers genetiske forandringer. Metoden har s\u00e5ledes potentiale til i nogen situationer at overfl\u00f8digg\u00f8re tumorbiopsiere, idet man kan f\u00e5 den molekyler genetiske diagnose fra en blodpr\u00f8ve.<\/p>\n

Regulatory mechanisms in Diffuse Infiltrating Pons Gliomas (DIPG)<\/h2><\/div>

Research field: In Vitro models (cell cultures), RNA regulation in DIPG, epigenetic regulation, screening models for drugs. Pilot studies for the establishment of Patient derived xenograft (mouse models).<\/h4><\/div>

Status:
\n1) Functional analysis of DNA damage signalling pathways and selected DNA repair pathway in paediatric H3K27M mutant and H3K27 wild-type gliomas has been performed. Further investigation of the activation of DDR with and without ionising radiation needs to be included as well as a microscopy-based quantification of foci formation to see the broader picture.<\/h4><\/div>

2)
\nA drug screening using the CNS-permeable drug library for radiosensitivity of H3K27M-mutated paediatric gliomas has been performed. This objective was split into two sub-objectives A (CNS drug screen) and B (drug validation and in vivo assessment).
